Bacterial Community Structure of Rhizosphere Soil of Cynanchum bungei Decne Based on High-throughput Analysis

【Objective】The study was conducted to understand the structure and composition of rhizosphere soil microbial community of Cynanchum bungei Decne, which provided theoretical basis and scientific support for making full use of rhizosphere microbial resources, optimizing rhizosphere growth-promoting bacteria and improving imitation wild planting technology.【Method】With Illumina MiSeq high-throughput sequencing technology, the richness and diversity of rhizosphere soil bacterial communities of Cynanchum bungei Decne were analyzed and determined.【Result】According to the sequencing results, a total of 5 404 OTUs were obtained from 6 rhizosphere soil samples, belonging to 22 phyla, 63 classes, 98 orders, 170 families and 274 genera, among which the dominant phyla of LW20 was Acidobacteria, with a relative abundance of 24.76%; the dominant phyla of LW10, LX10, LX20, CQ10 and CQ20 were Proteobacteria, with a relative abundance of 57.89%, 46.00%, 38.22%, 27.29% and 31.05%, respectively; the dominant genus of LW10 was Pseudomonas, with a relative abundance of 22.35%; the dominant genus of sample LW20 was Spartobacteria general incertae sedis, and the relative abundance was 21.37%; the dominant genus of sample LX10 was Solirubrobacter, and the relative abundance was 5.57%; the dominant genus of LX20 was Sphingomonas, and the relative abundance was 4.31%; the dominant genus of CQ10 was Norank acidobacteria GP4, and the relative abundance was 9.58%; the dominant genus of sample CQ20 was Streptophyta, and the relative abundance was 9.02%【Conclusion】Comprehensive comparison showed that the bacterial community abundance and diversity of sample CQ10 were the highest, and Chao and Ace indexes were 1 150.41 and 1 102.51, respectively. The bacterial community distribution of sample LX10 was the most uniform, and Shannoneven index was 0.8315. With the deepening of underground rhizome depth, the abundance and diversity of bacterial microbial community decreased. There were significant differences in soil bacterial communities among samples from different producing areas and rhizome depths.
