Autologous platelet rich plasma have positive eff ect on ram spermatozoa during cryopreservation in non-breeding season

The aim of this study was to evaluate the eff ects of autologous platelet-rich plasma (PRP) supplemented egg yolk based extender on ram semen after thawing. Semen samples were collected from nine Kıvırcık rams and each semen was divided into four equal aliquots and mixed with diff erent concentrations of PRP supported extenders [no PRP (control), 12.5x106/mL PRP, 25x106/mL PRP, or 50x106/mL PRP)]. Motility, plasma membrane functional integrity, acrosome integrity, mitochondrial membrane potential, DNA integrity and malondialdehyde concentrations (MDA) were measured and analyzed after thawing. The results showed that 25x106/mL PRP group had positive eff ect on motility (61.67±3.81), membrane functional integrity (71.00±2.96) and MDA levels (5.13±0.64) at post-thawed (P<0.05). It was determined that 25x106/mL PRP and 50x106/mL PRP groups were more eff ective than other groups in terms of mitochondrial membrane potential (69.50±1.93), acrosome integrity (78.04±2.65) and DNA integrity (5.33±0.92). The results of the study showed that autologous PRP has a protective eff ect in the cryopreservation of ram spermatozoa.
