Protocol for delivering proteins, peptides, and shRNAs via the intrathecal route in the experimental allergic encephalomyelitis mouse model
Abstract
Summary: The blood-brain barrier hinders drug delivery to the central nervous system (CNS), particularly for large molecules. Here, we present a protocol for delivering proteins, peptides, and short hairpin RNAs (shRNAs) via the intrathecal (IT) route in the experimental allergic encephalomyelitis (EAE) mouse model. We describe steps for developing EAE in mice and administering treatments intrathecally. The insights into treatment efficacy that can be provided by this protocol make it an important tool for CNS research. For complete details on the use and execution of this protocol, please refer to Bhusal et al.1 : Publisher’s note: Undertaking any experimental protocol requires adherence to local institutional guidelines for laboratory safety and ethics.
