Summary: Laser capture microdissection (LCM) coupled with transcriptome profiling is a powerful technique that allows for tissue-specific gene expression analysis in a complex system. One major challenge in using this technique is to obtain RNA without compromising its integrity. Here, we present a protocol optimized for radish root tissue sections using Steedman's wax embedding to obtain high-quality RNA suitable for next-generation sequencing analysis.For complete details on the use and execution of this protocol, please refer to Hoang et al.
