Tanigawa et al. investigate the role of Pib2, a vacuolar protein, in the glutamine-dependent upstream regulation of TORC1 in budding yeast. Their study reveals that Pib2 plays dual roles as a glutamine sensor and a direct TORC1 activator, a finding useful for the community working on TORC1 upstream regulation.
