The effect Mesenchymal stem cell treated with 17-β estradiol on the future of the innate immunity responses of Rheumatoid Arthritis induced with collagen in Wistar rats
Abstract
Background: This study was done to investigate the effect of 17-β estradiol (E2) treatment on the potential of Mesenchymal stem cells (MSC) for modulation of innate immunity responses in the animal model of Rheumatoid arthritis (RA). Methods: After isolation of MSCs, they were co-cultured with 100nµM 17-β estradiol for 24 h. RA was induced by collagen peptide and complete Freund's adjuvant in Wistar rats. One week after immunization, rats were allocated in groups received untreated MSCs (2×106 cells-intraperitoneal), received MSCs pulsed with estradiol and untreated rats. The change in the diameter of wrists and ankles of each rat was recorded every 5 days until 33 days after induction. Results: The edema and swelling of ankles in the positive control group (5.9 ± 0.6mm) was higher than the treatment groups. foot swelling in RA rats received MSCs treated with 17-β estradiol (1.69±0.45mm) were significantly decreased (P<0.05) compared to RA rats received MSCs alone (2.96±0.3 mm). Also, the rats of respiratory burst (1.57 ± 0.11), phagocytosis rate of neutral red (1.11 ± 0.09) and nitric oxide production (219.7± 16.81µM) in splenocyte population of the positive group were higher than treatment groups. The levels of respiratory burst, phagocytosis rate and nitric oxide in RA rats received MSCs pulsed wit estradiol were (0.79 ± 0.09, 0.61 ± 0.08 and 155.9 ± 13.29 µM), respectively. These factors in the RA rats received MSCs were (0.99 ± 0.08, 0.88 ± 0.07 and 187.9 ± 12.18 µM), respectively. Conclusion: 17-β estradiol-treated MSCs increases regulatory function and inhibitory effect on the inflammatory mediators of spleen innate immune cells in RA model compared to un-treated MSCs.
