Effect of Microstructure of Micro-Arc Oxidation TiO2 Coating on Cell Adhesion Properties

For improving the biocompatibility of TA2 pure titanium surface,micro-arc oxidation technology was used to prepare four kinds of titanium oxide coatings with different microstructures on TA2 pure titanium substrate by setting different electrolytes and electrical parameters.XRD, SEM, step profiler, and contact angle measuring instrument were employed to analyze the phase composition, micro morphology, roughness and hydrophilicity of coatings.Furthermore,MC3T3-E1 (mouse osteoblasts) were co-cultured with the titanium oxide coatings with different microstructures to observe the adhesion status between cells and the surface of the sample.Results showed that titanium oxide coatings with structural morphology of smooth macropores and rough macropores could be prepared in phosphate system electrolyte.By observing the adhesion state of cells using SEM, it was found that filopodia were generated on the surface of titanium oxide coatings with these two structural morphology, and cell secretions could be seen on the surface, indicating that the coating surface had certain biological activity.Moreover, the titanium oxide coatings with structural morphology of smooth pores and small rough pores were prepared in NaOH system electrolyte.On the surface of the titanium oxide coating with these two structural morphology, the adhesion area of cells was significantly increased, lamellipodia and filopodia were abundant, and the biological activity was greatly enhanced.The further comparative analysis suggested that the titanium oxide coating could guide the migration direction of cells when the roughness was high, and could improve the adhesion ability of the coating surface to cells when the roughness was low.Besides,the coatings with micro and nano dual-order structure had an integration effect on the two kinds of abilities.
