Histomorphometric study of human thyroid follicles with digital holographic microscopy

Introduction: Digital holographic microscopy has allowed optical microscopy to make use of numerical and computational tools, making multiple contributions to the study of cells and tissues in different fields of medicine and other related sciences. Objective: Describe the histological and morphometric characteristics of human thyroid follicles with digital holographic microscopy. Method: A descriptive and cross-sectional study of human thyroid follicles was performed, from the histomorphometric point of view, using a digital holographic microscopy facility. The paraffin inclusion technique, and hematoxylin-eosin staining was used for sample processing. From 10 to 12 hologram captures were taken per sample and the double propagation method for hologram reconstruction. The area, perimeter, major and minor diameters of follicles and follicular cavities were obtained and holographic reconstructions in three dimensions were performed. The arithmetic mean was measured as a central tendency measure and the standard or standard deviation as a measure of dispersion. Results: Follicular parameters: Area (5 140.31 ± 1 126.71 µm2); Perimeter: (2 961.54 ± 71.2 µm); Larger diameters :( 921.17 ± 24.34 µm) and smaller: (746.67 ± 18.08 µm); Epithelium height: (7.92 ± 0.96). Follicular cavities: Area: (3 686.18 ± 1,023.52 µm2); Larger diameter: (698.86 ± 19.55 µm) and smaller: (581.15 ± 13.82 µm). Conclusions: There are follicular parameters, determined by digital holographic microscopy, not reported by the literature consulted that are of interest in the histological study of human thyroid follicles.
