Abstract

The purpose of cytotoxicity test is to assess the toxic effect of a substance directly to the tissue culture. MTT assay is an assay on microplate which need no cell transfers. The method may be used for evaluating proliferation and cytotoxicity of a substance on cells in a quick, sensitive, and semiautomative manner without radioactive isotop. The assay is based on the ability of living cells to reduce the yellow soluble 3-(4,5-dimethylthiazol-2-y1)-2,5-di-phenultetrazolium bromide (MTT) salts to purple-blue insoluble formazan which involves the enzymes of endoplasmic reticulum and mitochondria. The number of viable cells is therefore measurable as the concentration of the solubilized formazan measured by a spectrophotometer.
