Study on separation, purification and chemical structure of Curdlan polysaccharide
Abstract
Curdlan polysaccharide was isolated from the fermentation broth of Alcaligenes faecelis var. C125 by acid and alkali and the rude sample CD-1 was gained. The protein and pigment were removed from CD-I by Sevage method and cellulose column chromatography, respectively. The pure sample CD-2 was separated by Sephadex G-100 column chromatography. The CD-2 was a homogeneous pure polysaccharide confirmed by UV-spectrometer and Sephadex G-200 chromatography, and its molecular weight determined by Sephtrometer-200 column chromatography was 65300D. CD-2 was composed of only glucose, which was proved by paper chromatography analysis after complete hydrolysis of the pure sample by sulfuric acid. And the chemical structure of CD-2 was indicated as a kind of linear glucan linked by β- (1, 3) linkage by infrared spectroscopy and 13C-NMR.
