Expression of miR-409-3p in ovarian serous carcinoma and its effect on biological behaviors of HO-8910 cells

Objective To investigate the expression of miR-409-3p in ovarian serous carcinoma and its correlation with the clinicopathological characteristics of the patients and observe the effect of miR-409-3p overexpression on the proliferation, invasion and apoptosis of HO-8910 cells. Methods We collected 40 tissue specimens of ovarian serous carcinomas and 20 normal ovarian tissues from patients undergoing surgeries for ovarian serous carcinoma in our hospital between January, 2014 and December, 2014. We examined the expression of miR-409-3p in the tissue specimens using RT-qPCR and explored the correlation of miR-409-3p with the clinicopathological parameters of the patients. We also detected the expression of miR-409-3p in 2 ovarian serous carcinoma cell lines (HO-8910 and SKOV3) and a human ovarian epithelial cell line (IOSE80). The effects of transient transfection with miR-409-3p mimics on the proliferation, apoptosis and invasion of HO-8910 cells were observed using CCK8 assay, flow cytometry and Transwell invasion assay, respectively. Results The expression of miR-409-3p was significantly lower in ovarian serous carcinoma tissues than in normal ovarian tissues (P 0.05). The patients with low miR-409-3p expressions had significantly shorter overall survival than those with high miR-409-3p expression levels (P < 0.05). The expression of miR-409-3p was also significantly lower in HO-8910 and SKOV3 cells than in IOSE80 cells, and overexpression of miR-409-3p significantly suppressed the proliferation and invasion and promoted the apoptosis of HO-8910 cells (P < 0.05). Conclusion miR-409-3p is lowly expressed in ovarian serous carcinoma and its expression level is significantly correlated with the FIGO stage, omental metastasis and overall survival of the patients. Overexpression of miR-409-3p inhibits the proliferation and invasion and promotes apoptosis of HO-8910 cells, suggesting the value of miR-409-3p as a potential prognostic indicator and a therapeutic target for ovarian serous carcinomas.
