Potential use of anti-cancer drugs coated scaffolds for local drug delivery in cancer cell lines
Abstract
Objectives: To optimize and characterize anticancer drugs conjugated biodegradable scaffold material. To evaluate drug release profile of anticancer drugs conjugated biodegradable scaffold material. To evaluate the effect of scaffold mediated localized chemotherapy on cancer cell lines including oral, breast and prostate cancer in terms of apoptosis assay, proliferation assay and gene expression analysis. To analyse the results for drug release profile and effectiveness of local drug delivery on different cell lines. Methodology: A lyophilized hydrogel of medium molecular weight chitosan, anticancer drugs (in three different concentrations viz; 0.5 μM, 1 μM and 1.5 μM) and sodium triphosphate is prepared to evaluate drug release profile. Efficacy of anticancer drugs was compared when used in its pure and free form with when it was used along with scaffold in same dosage on three cancer cell lines viz; oral, breast and prostate cancer. Results: Dose-dependent decrease in cell proliferation and increase in cell apoptosis was seen in all three cancer cell lines. Conclusion: SMLC is effective in increasing the bioavailability of drug at localised site and reducing systemic side-effects of anti-cancer drugs.
