Trachycephalus mambaiensis was recently described from two localities in the State of Goiás, in central Brazil. In this paper a new record of T. mambaiensis is presented, which is the first one in the State of Minas Gerais, Brazil. The species was found in three localities in the Municipality of João Pinheiro, a Cerrado area in northwest Minas Gerais.
