The development of nanomedicine is increasing significantly nowadays due to its effectless and efficiency than before. One recent nanomedicine innovation is molecule gates material mesoporous silica that supports drug delivery systems. The gates could be opened or closed in response to an external or internal command or stimulus. The internal stimulus consists of pH, redox, and enzymes. In contrast, the external stimulus consists of light and thermal stimuli. The gates can respond to the stimulus. As a result, the drug will be released. To modify nano gates, the materials and the drug are crucial for the design and functionality of these systems and can be effective in achieving the goal. This review explains the molecule-gated mesoporous silica nanomaterial (MSNs), the mechanism of internal/external stimulus of the nano gates in drug delivery systems (DDS), the potential and the biocompatibility of mesoporous silica nanomaterial (MSNs) as a nano gate and the pathways or methods to modified them.
