Bukti Baru Infeksi Natural Wolbachia sp. pada Aedes aegypti dengan Aedes albopictus dari Makassar

Abstract There is currently no research on the presence or absence of Wolbachia bacteria naturally in Ae. aegypti and Ae. albopictus mosquitoes in Makassar City. Therefore, it is necessary to study "New Evidence of Natural Presence of Wolbachia sp. in Ae. aegypti and Ae. albopictus from Makassar". This research was an explorative and analytic study with a cross-sectional study approach. The sampling of mosquitos was carried out in the Panakkukang District and Biringkanaya District of Makassar City from October to December 2018. The methods used were the Light trap method, Resting collection, and Larva collection. After that, the morphological identification process of Ae. aegypti and Ae. albopictus was carried out microscopically. Then extraction and purification of Wolbachia DNA using the phenol-chloroform method were used and continued by detecting the wsp gene using PCR and electrophoresis. From 33 Ae. aegypti mosquito pool samples in Makassar City, 28 pools were found from the middle of the city and 5 pools from the suburbs, while from 4 Ae. albopictus mosquito pool samples were found 1 pool from the middle of the city and 3 sample pools from the suburbs. Wolbachia infection was found naturally in Ae. aegypti mosquitoes (2 pools) as in Ae. albopictus mosquitoes (2 pools) in Makassar City. Abstrak Saat ini belum ada penelitian tentang ada tidaknya bakteri Wolbachia secara alamiah pada nyamuk Ae. aegypti dan Ae. albopictus di Kota Makassar. Oleh karena itu, perlu studi mengenai “Bukti Baru Infeksi Natural Wolbachia sp. pada Nyamuk Ae. aegypti dan Ae. albopictus di Makassar”. Penelitian ini merupakan penelitian eksploratif dan analitik dengan pendekatan cross sectional study. Pengambilan sampel nyamuk dilakukan di Kecamatan Panakkukang dan Kecamatan Biringkanaya Kota Makassar selama periode Oktober–Desember 2018 dengan metode light trap, resting collection, dan koleksi jentik. Setelah itu dilakukan proses identifikasi morfologi Ae. aegypti dan Ae. albopictus secara mikroskopis, kemudian ekstraksi dan pemurnian DNA Wolbachia menggunakan metode phenol-chloroform, dilanjutkan dengan mendeteksi gen wsp menggunakan teknik PCR dan elektroforesis. Dari 33 pool sampel nyamuk Ae. aegypti di Kota Makassar ditemukan 28 pool dari tengah kota dan 5 pool dari pinggiran kota, sedangkan dari 4 pool sampel nyamuk Ae. albopictus ditemukan 1 pool dari tengah kota dan 3 pool sampel dari pinggiran kota. Ditemukan infeksi Wolbachia secara natural pada nyamuk Ae. aegypti (2 pool) sebagaimana pada nyamuk Ae. albopictus (2 pool) di Kota Makassar.
