The development of quantative and qualitive analysis methods of suppositories with Maclura Pomifera extract

Chronic prostatitis and BPH are still very common diseases. In recent years, herbal preparations are widely used in the treatment of prostate diseases gland. The effectiveness of herbal medicinal products derived from MacluraPomifera is associated with their content of phytosterols and terpenes. Derived oil extract of MacluraPomiferafruit Orange (Maclurapomifera, Moraceae) is a rich source of terpenes and phytosterols. Previous studies indicated, that the content of such substances as lupeol and β-sitosterol, which is known its prostatoprotectors properties as well as the presence of isoflavones possessing anti-inflammatory and antioxidant properties. Aim of the work The aim of this work is a developing of methods that allow assaying qualitative and quantitative assessment of the suppositories with MacluraPomifera extract. Materials and methods Theobjects of this study are the suppositories with oil extract of MacluraPomifera. For the suppositories ingredients’identification thin layer chromatography has been used. Quantitative determination of active compounds has been carried out by spectrophotometry in ultraviolet and visible spectrum. The spectrophotometers of Thermo Scientific Evolution S60 (USA) and Apel PD303S (Japan) have been used. Determination of phytosterols and triterpenes amount has been performed in the equivalent of a reliable sample of lupeol ('Santa Cruz Biotechnology', USA; CAS: 545-47-1). Determination of isoflavones amount has been carried out in the equivalent of a reliable sample osayin ('BioBioPhaCo., Ltd.',China; CAS: 482-53-1). Results and discussion For identification of phytosterols and isoflavones in the suppositories composition a method of identifying their joint presence by TLC has been developed. Inalcoholic extraction from suppository mass two purple spots with Rf 0,8 and 0,57 are observed at the level of spots solution (lupeol and β-sitosterol) and two yellow spots with Rf 0,45 and 0,21 are observed at the level of spots solution comparison (osain and pomiferin). For the development of the quantitative determination method fortriterpenes and phytosterols amount in the suppositories we used their ability to move in the non-polar solvents from unsaponifiable fraction, and further to give colored complexes by interreaction with concentrated sulfuric acid. The absorbency of the derived colored solution has been determined at spectrophotometer at a wavelength of 309 nm. The method of differential spectrophotometry based on the selective interaction of aluminum chloride solution with the compounds of the flavonoid structure has been used for the quantitative determination of the isoflavones amount. Absorbance was measured in the visible spectrum at 419 nm. Thus, we have developed the methods that allow making qualitative and quantitative assessment of suppositories with MacluraPomifera extract. Relative mistake did not exceed ± 3,13% for the amount of phytosterols and triterpenes, and ± 2,24% for the amount of isoflavones.
