NLRP3 inflammasome: the core mechanism and potential target of inflammatory response in Alzheimer's disease

Alzheimer's disease (AD) is the most common cause of cognitive impairment in the elderly. However, the pathogenesis of AD remains unclear. Persistent excessive inflammatory response plays a pivotal role in the development of AD. It has been suggested that activation of the nucleotide-binding oligomerization domain⁃like receptor protein 3 (NLRP3) inflammasome is important for inflammatory response in AD. This article reviews the structure and activation of NLRP3 inflammasome and the specific inhibitors of NLRP3 inflammasome. DOI:10.3969/j.issn.1672⁃6731.2020.01.006
