Controllable loading and release of nanodrugs in polymeric vesicles

Nanomedicines, mainly consisting of liposome nanodrugs, polymer nanodrugs, protein nanodrugs, drug nanocrystals and inorganic nanodrugs, usually show higher efficiency than conventional small molecular drugs. Polymeric vesicles, owing to the advantages of good stability, easy functionalization, and large-scale production, have received extensive attention in drug loading and release. The hydrophilic cavity and hydrophobic vesicle wall can simultaneously encapsulate hydrophilic and hydrophobic drugs, which was beneficial to exert the synergistic therapeutic effect of drugs. In this review, we focused on the loading and release of nanodrugs in polymeric vesicles. The application of polymeric vesicles for single, multi-drug loading and co-assembly of polymers and nanoparticles were reviewed. The research progress of polymeric vesicles in the fields of targeted transport, drug release kinetics and stimuli-responsive controlled release of drugs was discussed.
