Regulation of insulin sensitivity in adipocytes by ZFYVE28

ZFYVE28, a FYVE domain-containing protein, is implicated in the regulation of endosomal trafficking and has been shown to influence insulin receptor degradation. While its role has been characterized in liver cells, its impact on adipocyte insulin sensitivity warrants further exploration. This study investigates the regulatory function of ZFYVE28 in adipocyte insulin signaling, elucidating its role in the degradation of phosphorylated insulin receptors through enhanced late endosome production. Using cell culture, gene expression manipulation, RNA and protein extraction, RT-qPCR, and Western blotting, we demonstrate that ZFYVE28 negatively regulates insulin sensitivity in adipocytes. Overexpression of ZFYVE28 results in decreased levels of phosphorylated insulin receptors, leading to inhibited insulin signaling pathways, as evidenced by reduced phosphorylation of ERK and AKT. Conversely, knockdown of ZFYVE28 significantly enhances these signaling parameters, suggesting its potential as a therapeutic target for ameliorating insulin resistance. Our findings not only extend the understanding of ZFYVE28’s cellular roles but also highlight its potential as a novel intervention point for metabolic disorders related to insulin resistance.
