Novel nano-leveled green switch-off fluorimetric technique for the determination of fexofenadine drug using Cilefa Pink B, a biological dye; raw material, dosage forms, and in vitro application; system kinetic study
Abstract
As a second-generation antihistamine, fexofenadine hydrochloride is prescribed to patients with allergic rhinitis and chronic urticaria to relieve their allergy symptoms. This article introduces the first novel, an efficient and eco-friendly spectrofluorimetric method for analyzing the fexofenadine anti-itching agent. Fexofenadine was measured and validated using a green, one-pot, and direct spectrofluorimetric approach in this study. Fexofenadine and Cilefa Pink B reagent in a slightly acidic solution were conceived to create an instant association complex. On the basis of off-effect fexofenadine's on the emission amplitude of the biological dye (Cilefa Pink B) at 554.5 nm, the fluorometric analysis was carried out. Linearity is between 0.1 and 2.0 µg mL−1, with sensitivity limits of 0.024 and 0.073 µg mL−1. Fexofenadine-Cilefa Pink B complexes were modulated analytically for all system parameters. It was also determined that the system met ICH standards. The proposed model also achieved a high recovery rate for fexofenadine in prescribed pharmaceutical dosage forms. Besides, the developed fluorimetric system was successfully used to track the drug of interest in human biofluids. The yielded product complex was characterized and confirmed by the FTIR technique. Additionally, the research was also done on the kinetics of the reaction system. Finally, the proposed method was assessed regarding its environmental friendliness using eco-scale.
