Objective To observe the effect of gender on animal model in phenylhydrazine(PHZ)-induced hemolytic anemia(HA). Methods Ten to 11 week old C57BL/6 mice were injected intra-peritoneal with PHZ solution (120 mg/kg). Peripheral blood was collected from submandibular vein before PHZ injection and on the 3rd, 5th, 7th, 9th and 14th days after PHZ injection. The indexes of red blood cells and white blood cells were detected by auto hematology analyzer. Results On the third day after PHZ injection, both female and male mice showed similar symptoms of acute HA: pale paw skin; hemoglobinuria; weight loss; decreased erythrocyte counting and hematocrit(P<0.05). On the third day after PHZ injection, the compensatory increase of spleen size in female mice was significantly higher than that in male mice(P<0.05). On the 9th day after PHZ injection, erythrocytes number, hematocrit and hemoglobin content of female mice were also significantly higher than that of male mice(P<0.05). Conclusions There are sex differences in stress erythropoiesis induced by PHZ in mice. The recovery rate of erythropoiesis in female mice is higher than that in male mice.
