Development of ACRS-PCR Method for Detection of Single Nucleotide Polymorphism g.66493737C/T of the Equine Myostanin Gene (MSTN)

Myostatin, encoded by the MSTN gene, is a member of the TGF-β super family that regulates 33 skeletal muscle development in a range of mammalian species including the horse. The aim of this work was designed of molecular-genetic method for detection of single nucleotide polymorphism g.66493737C/T of the equine myostatine gene (MSTN) in the breeds of horses Slovak warmblood and Norik and analysis of genotype. The SNP polymorphism C/T of the MSTN gene was studied in a group of 63 animals (50 Noriker horses and 13 Slovak warmblood horses). Genomic DNA was isolated from samples of hairs by commercial column kit QIAamp DNA Mini kit. The SNP g.66493737C/T was analyzed by ACRS-PCR method with restriction endonuclease RsaI. The allele C was detected by two restriction fragments 139 bp and 28 bp and the allele T with one 167 bp fragment. In the analyzed population of horses were detected the following frequency of alleles and genotypes for the SNP polymorphisms C/T of the MSTN gene. Frequencies of allele T and allele C were 0.8095 and 0.1905 and frequencies of genotypes were 0.0159 (genotype CC), 0.3492 (genotype CT) and 0.6349 (genotype TT).
