Preparation of Brucella antigen for diagnosis of Brucellosis in Khorasan

Objective: In this study, the antigen of Brucella was prepared. This antigen is applied for detection of Brucellosis through Wright test. Methods: This antigen can be used in every low equipped laboratory for Wright test. After continuous reviewing of Iranian and foreign references, Brucella abortus (Strain 19) were cultured and propagated and then, a suspension of bacteria (In normal saline containing phenol 0.5%) was prepared in sterile conditions and centrifuged. We added the solvent to that and standardize it with spectropherometric method in a wave length of 650nm. Findings: In comparison between commercial antigen preparation of Pasteur institute of Tehran and freshly prepared antigens by this method on 500 samples of serum gained from patient suspected to Brucellosis, Wright test was negative for 427 patients, but the rest of the patients (73) detected to be positive both with standard and freshly prepared antigen. These results wee acceptable and unquestionable agglutination was observed. Conclusion: It is important to remind that this antigen can be prepared for daily applications and its preparation possible in any laboratory even with minimum equipment.
