Poor hematopoietic function is the common feature of bone marrow(BM) failure diseases. However, the pathogenesis of BM failure diseases has not been fully clarified. Under physiological conditions, the self-renewal and fate determination of hematopoietic stem cells are strictly regulated by BM microenvironment. Hematopoietic stem cell transplantation is not only an effective way to treat leukemia, but also an ideal model to study hematopoietic regulation. Moreover, poor hematopoietic reconstitution after transplantation is a good disease modelfor the study of BM failure diseases. Taken together, the successful implementation of translational studies based onnovel mechanisms may provide novel therapeutic strategies for patients with BM failure diseases in the future.
