Screening and identification of β-glucosidase-producing strain and enzymatic properties of crude enzyme
Abstract
β-glucosidase-producing strains were selected from the soil of decayed trees in primitive forests, and the β-glucosidase production ability was measured. The morphological characteristics, molecular biological identification and enzymatic properties of the high-yield strains were studied. The results showed that 13 β-glucosidase-producing strains were screened, a high-yield glucosidase strain PX14A was obtained through re-screening, and the β-glucosidase activity was 4.9 U/ml. The strain PX14A was identified as Bacillus subtilis. The results of enzymatic properties research showed that the β-glucosidase of the strain PX14A crude solution was stable below 65 ℃, the optimal reaction temperature was 60 ℃, the enzyme was stable at pH 4.0-7.5, and the optimal reaction pH value was 6.5. Mg2+ and Fe2+ with a concentration of 1 mmol/L had a good activating effect on enzyme activity. The results indicated that the strain PX14A had good application prospects.
