Using alternative absorption path for vinpocetine, in particular rectal, will allow to improve its bioavailability, efficiency in comparison with traditional oral dosage forms and will provide practical cardiology with accessible domestic pharmaco-therapeutic product. On the department of technology of medication Zaporozhye State Medical University as a result of physical-chemical, biopharmaceutical and microbiological investigations vinpocetine rectal dosage form, namely suppositories with0,01 gof medicinal component on the lipophylic base (cacao oil with 0,5% surface-active substance twin 80) has been proposed. Consistent properties of suppository mass have direct influence on the technological parameters of rectal dosage forms manufacturing. Temperature is the most determinative for start of releasing, absorption and bioavailability of medicinal substances. The aim of this work is study structural-mechanical properties of vinpocetine rectal dosage form depending on temperature of suppository manufacturing technological process. Study of structural and mechanical characteristics of vinpocetine suppository mass on the cacao oil base with 2% of twin 80 has been carried out on the rotary viscosimeter «Reotest-2» with cylindrical arrangement by the body’s temperature37°Cand temperature of manufacturing technological process. Analysis of vinpocetine suppository mass on the cacao oil base with 2% of twin 80 rheogram by the37°Chas showed formation of hysteresis loop. That fact convincing proved the presence of restoring after destruction coagulation bonds in suppository mass structure. «Mechanical stability» of suppositories is 1,1. That fact also confirms high thixotropic properties of compositions and allows to provide complete restoration of structures after applying tension at the period of manufacturing technological process and even distribution of active substance in dosage form. Further investigations of rheological properties of vinpocetine suppository mass on the cacao oil base with 2% of twin 80 by the 500С showed a dilution of composition and maintenance of structured system properties. It is indicated by the hysteresis loop formation on the rheogram. Thixotropic properties dominance in the suppository mass confirms calculated insignificant difference of «mechanical stability» of composition 1,01. Revealed thixotropic properties of vinpocetine suppository mass on the cacao oil base by the 500С indicate even distribution of active substance in the suppository composition.
