Here the structure of the HEXIM arginine rich motif in complex with the stemloop-1 of 7SK snRNA is determined, and insights are provided into how the HIV protein Tat overcomes the structural constraints required to displace HEXIM, an event required for the extraction of pTEFb during transcriptional elongation.
