A new type of silicon-based bilayer photonic crystal slab biosensor is proposed based on the theoretical investigation of various parameters of two-dimensional photonic crystal slab on the peak wavelength and characteristics of the reflection spectrum. The spectral sensitivity of two-dimensional photonic crystal biosensors with different refractive index analytes and the response characteristic of these sensors were systematically analyzed using wavelength interrogation methods. The results show that selection of the appropriate lattice type (hexagonal lattice) and lattice constant (a = 275 nm) can improve the sensitivity of a bilayer photonic crystal slab biosensor. This analysis expands understanding of the biosensor design and its optical performance. Keywords: Photonic crystal slabs, Biosensor, Sensitivity, Hexagonal lattice
