Pendahuluan: Salah satu sumber senyawa bioaktif yang diyakini memiliki aktivitas antibakteri yang kuat adalah teripang. Teripang Timba Kolong (Holothuria sp.) merupakan spesies yang dominan di Kepulauan Mentawai Sumatera Barat Indonesia. Tujuan penelitian ini adalah untuk mengetahui aktivitas antibakteri teripang Timba Kolong yang ditemukan di Kepulauan Mentawai Sumatera Barat. Metode: Metode penelitian ini adalah laboratorium eksperimental. Aktivitas antibakteri teripang Timba Kolong diukur dengan menggunakan metode Kirby-Bauer dengan cara mengukur zona hambat terhadap Streptococcus viridans pada karies gigi. Absorbsi tiap sumuran diukur dengan spectrophotometer microplate reader pada panjang gelombang 570 nm. Uji aktivitas antibakteri dilakukan dengan post test only control group design dan metode disk difusion. Bakteri Streptococcus viridans dijadikan suspensi dengan konsentrasi 0,625; 1,25; 2,5; 5; 10 dan 20% dengan pelarut dimethil sulfoksida (DMSO). Hasil: Hampir semua sampel menunjukkan aktivitas antibakteri terhadap Streptococcus viridans dengan konsentrasi yang paling tinggi 0.625% yaitu 7.598 ± 0.8427 mm. Simpulan: Ekstrak Holothuria sp. memiliki aktivitas antibakteri terhadap pertumbuhan Streptococcus viridans. ABSTRACT Introduction: One of the natural sources with bioactive compounds that are believed to have strong antibacterial activity is the sea cucumber. Timba Kolong sea cucumber (Holothuria sp.) Is the dominant species found in the Mentawai Islands, West Sumatra, Indonesia. The purpose of this study was to determine the antibacterial activity of Timba Kolong sea cucumber found in Mentawai Islands, West Sumatra. Methods: This research method was an experimental laboratory. Antibacterial activity of Timba Kolong sea cucumber was measured using the Kirby-Bauer method by measuring the inhibitory zone of Streptococcus viridans in dental caries. The absorption of each well was measured by a microplate reader spectrophotometer at the wavelength of 570 nm. The antibacterial activity test was performed with a post test only control group design and disk diffusion method. Streptococcus viridans bacteria were made as suspensions with a concentration of 0.625; 1.25; 2.5; 5; 10; and 20% with dimethyl sulfoxide (DMSO) solvents. Results: Almost all samples showed antibacterial activity towards Streptococcus viridans with the highest concentration of 0.625%, which was 7.598 ± 0.8427 mm. Conclusion: Timba Kolong sea cucumber has antibacterial activity towards the growth of Streptococcus viridans.
