Detection of Cucumber Mosaic Virus (Cucumber Mosaic Virus, Cmv) Causing Natural Infection on Some Cultured Plants in Marmara Region

In this study, a total of 235 plant leaf samples were collected from 48 tomatoes from Çanakkale (Ezine), 15 tomatoes, 25 peppers and 35 gladiolus from Bursa (İznik, Yenişehir, Orhangazi), 12 cucumbers, 8 zucchini and 16 peppers from Yalova (Center), 25 peppers from Istanbul (Pendik), 14 tomatoes and 15 peppers from Bilecik (Osmaneli) and from 22 peppers from Sakarya (Pamukova) in June and July 2013. Samples were analysed by DAS-ELISA technique. 69 % of pepper samples, 87 % of tomato samples, 75 % of zucchini samples, 58 % of cucumbers sample and 69 % of gladiolus samples were found to be infected with Cucumber mosaic virus (CMV). Real-Time PCR analysis in the gladiolus, peppers, tomatoes, zucchini and cucumber samples revealed peak values between 84.0-88.7, while the negative control gave a peak value of 76.0. The expected amplicon sizes of 540 bp were observed after electrophoresis of PCR products in 1% agarose gel. The results of ELISA and PCR showed parallelism. This study is the first report of CMV in gladiolus in Turkey.
