THE RESEARCH OF BIOCOMPATIBILITY OF POLYMERIC MATERIALS (POLY-METHYL-METH-ACRYLATE AND BISPHENOL-A-DIGLYCEDYL-METH-ACRYLATE) IN A CORNEAL STROMA CELL CULTURE MODEL

Purpose. To evaluate a proliferative activity of cadaver donor corneal stromal cells (CSCs) in contact with the polymers poly-methyl-methacrylate (PMMA) and bisphenol-A-di-glycidyl-meth-acrylate (bis-GMA) in the cell culture (in vitro).Material and methods. The study was based on the method of cell culture of corneal stromal cells (CSCs) of cadaver donors under conditions of normothermia. Polymer samples served as objects of the research. There were 4 distinct modifications of bis-GMA, produced by the Reper-NN (Nizhniy Novgorod (the experimental groups K1-K4), and one of PMMA, produced by the ETP FEMI (Moscow)(the group K5). In the control group (the group K6) the polymer sample was not used. Cadaver donor CSCs suspension of the 7th passage was added over the samples (20000 cells in 2mL of culture medium per sample) and then cultured during 7 days within the wells of standard culture 24-well plates (a sample per well) in the standard normothermic conditions using the DMEM/F12 culture medium supplemented with fetal bovine serum and antibiotics mix. The culture medium was replaced every 3rd day. The main subject of the research was the CSCs proliferation degree evaluation. Cells were extracted from the 4 wells of each group daily and were counted. The Wilcoxon test was used for a statistical significance evaluation.Results. On the first day of observation no statistically significant difference in the level of proliferation degree between the groups was found. A statistically significant difference between the levels of keratocytes proliferation was revealed in the presence of test samples by the end of the observation period. Proliferation intensity rates were in order of the symptom diminishing as follows: (control, K5) >K1>(K3,K4)>K2. There was detected no significant difference between groups K3 and K4 (p>0.05).Conclusion. Among the investigated materials the polymerK2 had the lowest proliferation-inducing properties; K5 had the highest and others (K1, K3, K4) did not show an unambiguous tendency to the proliferation induction.
