In vivo real-time monitoring of anti-factor Xa level using a microdialysis-coupled microfluidic device
Abstract
The feasibility of in vivo real-time monitoring of anti-factor Xa level had been explored using an integrated microdialysis-coupled microfluidic system. By customizing the dialysis membrane and utilizing a fluorogenic substrate, the performance of the integrated device was characterized and tested by continuous monitoring of the anti-factor Xa level in rats. Efficient sampling for low molecular weight heparin in the blood stream was demonstrated and the detection range of the device was determined to be 0.1-1.5 IU/ml. The anti-factor Xa profile was recorded for a period of 7 hours. Moreover, the results generated from this method agreed well with those from conventional assay. The present system provides a novel quantitative monitoring method with great precision, good reproducibility and full data coverage of treatment period. This study has paved the road for developing a bedside drug monitoring device in hospital setting.
