LncRNA MYCNOS promotes prostate cancer cell proliferation by regulating the miR-466/RUNX2 axis

The function of lncRNA MYCNOS has only been reported in hepatocellular carcinoma. We studied the role of MYCNOS in prostate cancer (PC). In this study, MYCNOS expression in tissue samples from 64 PC patients was studied by RT-qPCR. Base pairing formed by MYCNOS and miR-466 were predicted by IntaRNA 2.0 and confirmed using dual luciferase activity assay. Overexpression of MYCNOS, miR-466, and RUNX2 was performed to explore their interactions. CCK-8 assay was applied to quantify cell proliferation. We found that MYCNOS was upregulated in PC. MYCNOS is predicted to interact with miR-466. However, MYCNOS and miR-466 did not regulate the expression of each other in overexpression experiments. Instead, MYCNOS increased RUNX2 expression, a target of miR-466. MYCNOS and RUNX2 overexpression resulted in increased PC cell proliferation, while miR-466 overexpression suppressed cell proliferation. In addition, MYCNOS reduced the inhibitory effect of miR-466 on cell proliferation. In conclusion, MYCNOS may promote PC cell proliferation by upregulating RUNX2 via sponging miR-466.
