Study on antioxidantactivity of Abelmoschus manihot (L.) medic polysaccharide on Caenorhabditis elegans

In order to study the biological roles of Abelmoschus manihot (L.) medic polysaccharide (AMP) in vivo and the underlying mechanism,C.elegans was used as a model to analyse the influence of ck(0 mg/mL),low dose group (25 mg/mL),medium dose group (50 mg/mL) and high dose group (100 mg/mL) AMP on physiological indexes,biochemical indexes and apoptosis of nematodes.The results show that AMP could prolong the life span of C.elegans,reduce the reproductive capacity,improve the motor capacity,lower the content of MDA and ROS,and increase the activities of SOD,CAT and GSH-Px under normal growth conditions.Under the condition of 200 μmol/L juglone oxidative stress,AMP can improve the survival rate of nematodes,effectively eliminate ROS,reduce the content of MDA,increase the activities of SOD,CAT and GSH-Px and inhibit cell apoptosis.AMP can enhance the resistance of C.elegans to oxidative stress by increasing the enzyme activities of antioxidant defense system and scavenging free radicals.AMP has the potential to be a new kind of natural polysaccharide with antioxidant activity.
