Effet du 2,4-D sur l’induction de l’embryogenèse somatique à partir de cotylédons matures de caroubier (Ceratonia siliqua L.)
Abstract
The present study is the first report of the production of embryogenic callus and structures cultures from mature cotyledons of Ceratonia siliqua. Regeneration depends on the plant material type and concentration of the auxin. In the case of mature cotyledons, the presence of 2,4 - dichlorophenoxyacetic acid was necessary for callus induction. This auxin promoted a rapid proliferated callus initiation with nodular structure in all explants and concentrations tested. Upon transfer to auxin free medium, the callus formed a large number of globular embryonic structures (SEG). The highest number of SEG (126/g of callus) was produced with 10 µM of 2,4-D. Higher concentrations were accompanied by hyperhydricity of SEG. No embryonic structures occurred from the others types of explants (immature seeds, leaves and portions of epicotyls, hypocotyls and roots).
