Isolation of Human Cumulus Granulosa Cells

INTRODUCTION: Follicles are the basic functional units of the ovary. It is characterized by the growth and development of ovarian follicles, proliferation and differentiation of granulosa cells. Oocytes are found in the graaf follicle surrounded by cumulus granulosa cells. Our aim in this study is to isolate the cumulus granulosa cells, which form the microenvironment of the oocyte, more easily and quickly. METHODS: Two female patients aged 25-30 years, with ten or more follicles and male factor infertility were included in the study. The cumulus granulosa-oocyte complexes obtained after OPU (oocyte pick-up) were denuded to separate them from each other. Then, the cumulus granulosa cells were seeded into culture dishes coated with Type I collagen and primary cell culture was established. RESULTS: Cumulus granulosa cells inoculated in flask culture dishes proliferated and became confluent on the 14th day. In the count made under the microscope, it was observed that 4x105 and 5x 105 cells were grown in the culture dishes, respectively. DISCUSSION AND CONCLUSION: The ability to culture cumulus granulosa cells more easily thanks to the isolation methods developed can accelerate the studies on obtaining quality oocytes and embryos.
