<em>Paeonias</em> sp. variety Sesu <em>in vitro</em> multiplication in Temporary Immersion Systems
Abstract
Paeonia are considered among the most important perennial ornamentals. The influence of three culture methods (semi-solid culture medium, static liquid culture medium and liquid culture medium by temporary immersion) as well as the type and concentration of cytokinins was investigated to improve the coefficients of in vitro multiplication of Paeonia sp. var. ‘SeSu’. Results demonstrated that temporary immersion increased significantly the multiplication coefficient by increasing the concentration of benzyladenine. A maximum concentration value of 6.66 mmol l-1 was reached. Both, type and concentration of cytokinins had effect on the multiplication of Paeonia var. SESu in its temporary immersion. The highest rates of multiplication were achieved with concentrations of 4.44 and 6.66 mmol l-1 meta-topolin and 6.66 mmol l-1 BA, without significant differences between them. Both cytokinins enhanced the shoot length. Hyperhydric shoots were not found. Key words: cytokinins, ornamentals, proliferation
