Mechanism of lncRNA JPX targeting miR-378 in tanshinone ⅡA affecting proliferation, cloning and apoptosis in colon cancer SW620 cells

Objective To study the mechanism of lncRNA JPX targeting miR-378 in the proliferation, cloning and apoptosis of colon cancer SW620 cells induced by tanshinone ⅡA. Methods Colon cancer SW620 cells were divided into control group, tanshinone ⅡA treatment group, vector group(transfected with negative control vector), JPX group(transfected with pcDNA-JPX), tanshinone ⅡA+vector group(transfected with negative control vector followed by tanshinoneⅡA treatment), tanshinoneⅡA+JPX group(transfected with pcDNA-JPX followed by tanshinoneⅡA treatment)), tanshinoneⅡA+JPX+miR-NC group(cotransfected with pcDNA-JPX and mimics control, followed by tanshinoneⅡA treatment), tanshinoneⅡA+ JPX+miR-378 group(cotransfected with pcDNA-JPX and miR-378 mimics, then treated with tanshinoneⅡA), tanshinoneⅡA+anti-NC group(transfected with inhibitor control followed by tanshinoneⅡA treatment), tanshinoneⅡA+anti-miR-378 group(transfected with miR-378 inhibitor followed by tanshinoneⅡA treatment). CCK-8 assay was used to detect cell proliferation, plate cloning to observe clone formation ability, and flow cytometry to measure cell apoptosis. Luciferase reporter system was adopted to study the targeting relationship between JPX and miR-378. Results Compared with the control group, the cell survival rate and number of formed cell clones were reduced, and the apoptotic rate was increased in the SW620 cells from the tanshinoneⅡA group(P<0.05). But the opposite results were seen when the JPX group was compared with the vector group(P<0.05), so were between the tanshinone ⅡA+JPX group and the tanshinone ⅡA+vector group(P<0.05), and between the tanshinone ⅡA+Anti-miR-378 group and the tanshinone ⅡA+Anti-NC group(P<0.05). The cell survival rate and number of formed cell clones were reduced, and the apoptotic rate was increased in the tanshinone ⅡA+Anti-miR-378 group than the tanshinone ⅡA+JPX+ miR-NC group(P<0.05). JPX targeted to promote miR-378 expression. Conclusion Tanshinone ⅡA induces apoptosis in colon cancer SW620 cells by down-regulating JPX targeting miR-378.
