Regulatory effects of a Grifola frondosa extract rich in pseudobaptigenin and cyanidin-3-O-xylosylrutinoside on glycolipid metabolism and the gut microbiota in high-fat diet-fed rats
Abstract
The aim of present study was to investigate the regulatory effects of an extract of the Grifola frondosa on glycolipid metabolism and the intestinal flora in high-fat diet fed rats. The ethyl acetate extract of G. frondosa (EAE), enriched with pseudobaptigenin and cyanidin 3-O-xylosylrutinoside, exerted a stronger inhibitory effect on lipase than the other polar fractions of G. frondosa in vitro. Supplementation with EAE (50 mg/kg) to rats for 8 weeks clearly ameliorated dyslipidaemia by suppressing the serum levels of glucose (Glu), total triglycerides (TG) and low-density lipoprotein cholesterol (LDL-C), and hepatic fat accumulation. Furthermore, EAE supplementation normalized the intestinal flora structure in high-fat diet fed rats. The relevance analysis suggested strong associations of Anaerotruncus, Barnesiella, Clostridiaceae, Erysipelotrichaceae, Paraprevotella, Clostridium_XVIII, Coprococcus and Turicibacter with the biochemical indexes. Simultaneously, the EAE treatment intervened the expression levels of the genes associated with glycolipid metabolism. Our results indicated that pseudobaptigenin and cyanidin-3-O-xylosylrutinoside can be used as pharmaceutical agents for the prevention of the disorder of glycolipid metabolism.
